What does tune to A440hz mean ... ?

[ A440Hz perfect pitch as is A1 55Hz but some keyboards don't go this low that is why we use 440Hz, also for tuning circuitry we use 400Hz linear to calibrate .

How can we measure harmonics... ?

[ With difficulty is the simple answer, for most musicians the easy way to reference a harmonic is to call it by its lay term either 3rd 5th or 7th.

Tuning methods.. ?

[ Most synths today have independent tuning built in so for each note you can choose your frequency association. Older synths modules are stuck in the European tuning mode, A# is exactly the same pitch as Bflat

]

Temperature tuning calibration... ?

[ To combat temperature the Asian tuning method is preferred.

Here you need to calibrate the # separate to the flat

So A# will have a different frequency to that of Bflat etc.
